Paul Peschisolido claimed the bragging rights against his former club as the Blades slipped to another pre-season defeat at the Pirelli Stadium.
The former Bramall Lane favourite saw his League Two side withstand second half pressure to cling to their lead.
Blades boss Kevin Blackwell included defensive trialist Gregory Vignal from Birmingham City, plus new signings Rob Kozluk and Jonny Ertl, in a strong starting line-up.
But Burton took the lead after Blades skipper Chris Morgan and central defensive partner Ertl had been caught out by a Jimmy Phillips cross.
Greg Pearson managed a six-yard shot past Unite's goalkeeper Mihkel Aksalu.
The Blades raised their tempo after the interval, but Burton's substitute goalkeeper, Kevin Poole, proved he had plenty offer on the eve of his 47th birthday.
Assistant manager Gary Speed said: "We thought we were the better team in the second half but Pooley at 47 was in good form and we just couldn't get it back."
